Abstract: This study investigates the effect of the addition of salt in the sample matrix on the separation efficiency of 2,3,7,8-TCDF and 2,3,7,8-TCDD using MEKC. The technique of using a high conductivity sample matrix in relation to the running buffer used is known as high conductivity sample stacking mode (HCSSM) and is carried out by varying the concentration of salt in the sample matrix. We varied the concentration of sodium chloride, NaCl (0-300 Mm) in the sample matrix and the optimized concentration of NaCl is 200 mM which provided the best separation in terms of peak intensity and efficiency. Thus the sensitivity enhancement factor obtained for the limit of detection (LOD) was 47.8 (TCDF) and 92.4 (TCDD) respectively.
